A Potential Use of Color Ultrasound as a Tool for Reproductive Management: New Observations Using Color Ultrasound Scanning that were not Possible with Imaging Only in Black and White OAK

Ultrasonography (US) has been applied to the ovary and the uterus of domestic animals from the late 1980s, and established in 1990s as a practical tool for animal production. US made it possible to detect pregnancy at a very early stage and, most importantly, to observe the real-time dynamics of follicular development and hence the discovery of follicular waves. This has greatly contributed to our understanding of ovarian physiology and helped us to develop several "pin-point" protocols for hormonal treatment. While US may not seem to fit preconceived ideas of a "green" technology, it does not contravene environmental priorities, and it is non-invasive ("ethical") and non-hormonal ("clean"). Using the US technology that is now commercially available at a...

Changes in Follicular Vascularity during the First Follicular Wave in Lactating Cows OAK
ACOSTA, Tomas J.; HAYASHI, Ken-Go; MATSUI, Motozumi; MIYAMOTO, Akio; 松井, 基純; 宮本, 明夫.
Increase in the blood supply to individual follicles appears to be associated with follicular growth rates and the ability to become the dominant follicle, while reduced thecal vascularity appears to be closely associated with follicular atresia. Therefore, this study aimed to determine the real-time changes in the vascularity of the follicle wall during the first follicular wave in cycling Holstein cows. Normally cycling and lactating cows (n=5) were examined by transrectal color Doppler ultrasonography (the sensitivity for velocity: > 2 mm/sec) to determine the changes in the vasculature of the follicle wall (presence or absence of blood flow) and the diameter of follicles. A new follicular wave and ovulation were induced by GnRH injection at 48 h...

Effect of intraluteal injection of endothelin type A receptor antagonist on PGF2alpha-induced luteolysis in the cow. OAK

Endothelin-1 (ET-1) is a luteolytic mediator in the bovine corpus luteum (CL), and its action appears to be via endothelin type A receptor (ETR-A). Thus, the aim of the present study was to determine the effect of ETR-A antagonist on PGF(2)alpha-induced luteolysis in the cow. Cows on days 1012 of the estrous cycle were subjected to five intraluteal injections of the ETR-A antagonist LU 135252 in saline or only saline at -0.5, 2, 4, 6, and 8 h after PGF(2)alpha administration (=0 h). Serial luteal biopsies were conducted to determine the expression of mRNA in the luteal tissue. There were no significant differences in the decrease in plasma progesterone (P) concentrations and the mRNA expressions of steroidogenic acute regulatory protein and 3...

Genital Blood Flow and Endometrial Gene Expression During the Preovulatory Period after Prostaglandin F2α-Induced Luteolysis in Different Luteal Phases in Cows OAK
JORDAN, Anne; HERZOG, Kathrin; ULBRICH, Susanne E; BEINDORFF, Nicola; HONNENS, Änne; KRÜGER, Lars; MIYAMOTO, Akio; BOLLWEIN, Heinrich; 宮本, 明夫.
The aim of this study was to examine the hypothesis that follicular and uterine perfusion as well as endometrial gene expression during the ovulatory period differs after induction of luteolysis during the 1st follicular wave compared with the 2nd wave or in intact cycle. Nine healthy non-lactating Holstein-Friesian cows were examined during three estrous cycles. A prostaglandin F2α analogue (PGF) was administered randomly either on Day 7 (1st wave cycle) or Day 11 (2nd wave cycle) after detection of ovulation (=Day 1). No hormonal treatment was used in the intact cycle with spontaneous ovulation. Transrectal Doppler sonography was conducted daily after PGF injection and in the intact cycle beginning on Day 18 of the estrous cycle until ovulation....

Ovulation of the preovulatory follicle originating from the first-wave dominant follicle leads to formation of an active corpus luteum OAK
MIURA, Ryotaro; HANEDA, Shingo; MATSUI, Motozumi; 羽田, 真悟; 松井, 基純.
The objective of our study was to compare the characteristics of the corpus luteum (CL) formed after ovulation of the dominant follicle (DF) of the first follicular wave (W1) and those of the CL formed after ovulation of the DF of the second (induced) follicular wave (W2). Non-lactating Holstein cows were used for this study. In Experiment 1, cows were treated with PGF2α and GnRH on days 6 and 8 (day 0 = day of follicular wave emergence) for W1 (n = 6) and W2 (n = 6), respectively. Dominant follicles were aspirated on day 9 to quantify the amounts of mRNA (VEGF120, VEGF164, FGF-2, StAR, P450-scc and 3β-HSD) in granulosa cells (GC). In Experiment 2, the size and blood flow area of the CL formed after ovulation of the DF in W1 (W1CL; n = 6) and W2 (W2CL; n =...

Subacute effects of a maximal exercise bout on endothelium-mediated vasodilation in healthy subjects BJMBR
Bousquet-Santos,K.; Soares,P.P.S.; Nóbrega,A.C.L..
We evaluated vascular reactivity after a maximal exercise test in order to determine whether the effect of exercise on the circulation persists even after interruption of the exercise. Eleven healthy sedentary volunteers (six women, age 28 ± 5 years) were evaluated before and after (10, 60, and 120 min) a maximal exercise test on a treadmill. Forearm blood flow (FBF) was measured by venous occlusion plethysmography before and during reactive hyperemia (RH). Baseline FBF, analyzed by the area under the curve, increased only at 10 min after exercise (P = 0.01). FBF in response to RH increased both at 10 and 60 min vs baseline (P = 0.004). Total excess flow for RH above baseline showed that vascular reactivity was increased up to 60 min after exercise (mean ±...
